Guerin was taken by the Knicks out of Iona with the 17th overall pick in the 1954 NBA draft. After a pair of seasons on active duty with the Marines, he made his NBA debut in 1956.

By his second season with the team, Guerin was an All-Star, averaging 16.5 points, 7.8 rebounds and 5.0 assists. It was his first of six consecutive All-Star selections, a run that included a trio of All-NBA second-team selections.

Guerin set a pair of then-team records with the Knicks. He set the franchise assist record with 21 in 1958; the total doubled as the Madison Square Garden record, and stood as the arena's best until John Stockton broke the record in 1999. And in 1959, Guerin scored 57 points against Syracuse, shattering the previous single-game Knicks record of 47 set by Carl Braun.

During the 1961-62 season, Guerin set three career bests: points per game (29.5, nearly eight points better than his previous high of 21.8), assists per game (6.9) and minutes per game (42.9).

New York made the playoffs just once during Guerin's seven years with the team, during the 1958-59 season. They were swept in two games in the Eastern Division semifinals by the Syracuse Nationals.

The Knicks dealt Guerin at the beginning of the 1963-64 season to the St. Louis Hawks for cash and a second-round pick, which ended up being Howard Komives.
